RATIONALE

A very good morning to honourable chief guest for today’s Program- Dr K. Manikya Kumari mam, HOD, Dept. of Botany, SJCW, Guest of Honour- Dr Rajesh Goud Gajula, Product Specialist (Molecular Biology), HIMEDIA, Sri Gopi krishna sir, Sr. Business Executive, Mr Leele krishna sir, Business Executive both from HIMEDIA, Respected Heads of the Departments, Faculties from SJCW and other colleges, participants from colleges and university and dear students of SJCW, Good morning to one and all.

This is Dr P. Mary Anupama, Convenor and Head of the Dept. of Biochemistry, SJCW extending a very warm welcome to the TWO-DAY NATIONAL WORKSHOP ON PCE BASED TECHNIQUES. 

RFLP and RAPD are essential molecular biology techniques used for detecting genetic variation. These techniques enable researchers and students to identify genetic differences among individuals or species.  Learning these methods provides a foundational understanding of genetic diversity, population structure, and evolutionary relationships, which are crucial in fields like agriculture, conservation biology, Forensic sciences  and genetic research.

Understanding these techniques can open up new research opportunities for students, helping them to analyse genetic data and apply molecular markers in breeding programs or biodiversity studies.

 Bacterial transformation is a key technique where students learn how foreign DNA (plasmid or linear) can be introduced into bacteria to express new genes. This process is fundamental in genetic engineering and biotechnology.  Practical experience with bacterial transformation helps participants gain confidence in handling microorganisms and understanding how genetic traits are manipulated by introducing genes for various applications, such as creating genetically modified organisms (GMOs) or producing bio-products like insulin.

Bacterial transformation is a cornerstone in the biotech industry, used in gene cloning, protein expression, and synthetic biology. The workshop would introduce participants to the basic principles behind these industrial applications, which are critical for careers in biotechnology, pharmaceuticals, and academic research.

Many students and early-career researchers are familiar with the theoretical aspects of molecular genetics but lack hands-on experience. This workshop offers practical, laboratory-based training in performing techniques like gel electrophoresis, DNA extraction, restriction digestion, PCR, and bacterial transformation, enhancing their lab skills and making them more competitive in academic and professional environments.

Workshops on these topics promote collaborative learning and provide opportunities for participants to have the Hand-on experience on the above-mentioned techniques empowering them with concepts and skills which keep them ahead of others.

LECTURE ON PCR AND ITS APPLICATIONS

The chief guest spoke on the principle and working of PCR. PCR is polymerase chain reaction which is used for amplification of DNA. This involves steps like denaturation , annealing and extension steps. The basic requirements for PCR are DNA template, primers, Nucleotides, Taq DNA polymerase and the equipment. The technique finds its application in southern blotting, northern blotting, finding out viral load, forenseic sciences, paternity testing, RFLP and RAPD. The workshop included the two topics RFLP and RAPD which are very important and hence the chief guest Dr Manikya kumari talked about them in detail.

Sir introduced the students with the principle, procedure of all the three experiments to be demonstrated in the workshop. immediately after that the hands-on experience of RFLP and RAPD has started. Himedia team with volunteers from the 3rd BBC have took up the entire 120 members into 5  batches and each batch was asked to come one after the other to handle the equipment and learn addition of reagents. First experiment was RAPD.

 Introduction to Transformation

Dr Mousami Shankar, Asst. Prof., Dept of Biochemistry has introduced the students with principle for transformation experiment.

Dr Mary Anupama, Convener has demonstrated the experiment and made students add plasmid and inoculate LB agar plates with transformed cells
